| Hi, I have a complicated question about reverse zymography. I read (and already tried) reverse zymography also works with proMMPs instead of the active form. This is quite logic as the proMMPs are activated in the gel via SDS causing the cystein switch. My question now is: what is the way of inhibition by TIMP then? as the prodomain is not cleaved in this activation mechanism,is the NTD of the MMP still available for the "normal" inhibitory binding of TIMP to MMP? |
