Post Bradford Assay Question

Hi everyone,

I will need to measure the total protein concentration in my samples.

If i am using bovine serum albumin (BSA) as the protein standard is it necessary to prepare the protein standard dilution using the lysis buffer that I used for my protein extraction?

Now i am using 0.1M sodium phosphate buffer (pH 6.8) and 1% Triton X-100 as my lysis buffers.

Does anybody have protocol for bradford assay where the O.D. values are measured by an ELISA plate reader and not spectrophotometer?

thanks
