Default protein running in the SDS gel

Hey All,

I am used to running a protein extract in an SDS gel at 4 degree celsius and used to get a high molecular protein band.

however i started running the same protein extract in a similar gel but at room temperature and didnt see any high molecular band.Although i could see a new band at a low molecular size.

Does anyone have come across such a case wherein, a high molecular wt protein band has moved down to a low molecular wt band just becoz of running of gel at room temperature?????

Hi
these is possibility that the protein may be hightly fragile but usually in SDS-PAGE temperature does not have much effect, but yes it has effect when you run Native-PAGE, but if somebody has experience it then please guide him.

Hello Maniche,
could be several possibilities. May be your proteins were precipitated to higher molecular weight complexes at 4C, and room temperature split those into lower molecular weight (by preventing protein precipitation).

may be a possibility.

Could you please fill us in on what you learn that is an interesting case.
