Default trouble with EMSA?

Hi,
At present, I am doing EMSA experiments with NF-kB, but I find it did not work although I have optimized the conditions. Any suggestions are welcomed. Thanks a lot.

"It doesn't work" isn't very descriptive. Do you mean that you can see probe on the gel but you can't see a shift when you add your protein/extract? Or that you get a shift but you can't supershift it with an antibody or see a change upon drug treatment? Or that you can't see your probe on the gel at all?
