Question comasie blue

please anyone reply quickly
Is there any risk if I leave the gel which have the protein in comassie blue stain for 2 or 3 days before destain
this will occur because the holiday of weakend

I would prefer to destain over the weekend, to avoid high background staining.

i think it wud give u problem just to destaining by long tim..otherwise u will get dark background of gel by which ur band will not be clearly visible...

It depends upon the concentration of the stain and the solvent being used. In my experience, higher concentrations of stain and the use of methanol/water solvents can lead to high background when the gel is stained for severeal days. In general, it is better to either stain for shorter periods of time (less than 24 hours) and then leave the gel in destain for several days, or to fix the gel first in 10% Acetic Acid for several days and then complete the staining/destaining when you return.
