Default total protein is high

I am working in identification of meat according to species by polyacrylamide gel electrophoresis
I worked for 4 month by this method :adding 0.9% Nacl to 2 gm meat and homogenize then centrifuge at 1400 RPM then take the supernatent at the same day
and inject the supernatent in the well of the gel by amount related to total protein of sample so I inject in the well about 5,6,15,20,10 microliter


at this month due to some reasons I left the hemogenate for 8 day then make centrifuge and extraction so when I measure the total protein it was very very high is it due to prolonged time the meat stay in saline or due to what?
and the amount injected in well is 0.9,0.8,0.6,1 microliter
so I inject 1 microliter in all sample because the fraction of microliter is very small


is there is any comment or explaination to my work
please anyone who work as me reply to communicate with him along my master degree

Homogenate in saline for 8 days and a large increase in total protein. Simple answer is that the tissue continued to disintegrate. Saline alone may not cause the cells to burst by osmotic pressure but the homogenization probably released lysozymes and proteinases.
