I am new to this forum and would like to ask for some advice Hi Everyone, I am new to the forum and need some advice, I am trying to prepare a grid screen with an automated instrument; I am preparing a 100mM HEPES pH gradient from pH 6.8 to 8.2 across 6 wells of a 24 well plate, What recommendation does anyone have for the appropriate base to get this gradient ? Running a TRIS gradient with the robot works great from pH 7 through 9 so I am confident of the instrument's ability but for the HEPES gradient, I cannot seem to find a suitable base to meet this, I tried a low NaOH solution but was concerned that I would dilute out the 100mM HEPES, Can anyone offer assistance ? I am more of an automation specialist rather than a biologist, thanks Automation101 |
