Default Tripeptide peptide extraction



I recently synthesized Phe-Phe-Arg using manual peptide synthesis. When I used Reagent R to cleave the peptide, addition cold ether did not yeild the precipitation of the peptide as expected. I researched a little and found that di- and tripeptides do not usually precipitate in cold ether but may form an oil. I rotovaped off the TFA remaining from the filtrate, however, the other components of reagent R have much higher boiling points. Can anyone lend a suggestion as to how to extract the peptide from the remains of Reagent R or if there is an alternative to Reagent R I can use that could be removed after peptide cleavage so that I can perform TFA/HCl exchange?

Hi

1) Please specify protecting group and resin you used for synthesis. Because If you used Arginine without protection then just treat peptide with dilute TFA without addition of scavanger, and then try to ppt out it with ether.

2) If you get sticky or oily product then distilled out Diethyl ether and try to use diisopropylehter to ppt out it again. if still get sticky solid decant out ether and lypholised to get soild.

try ether:n-pentane if not working. Just freezedry the oil in ACN:H2O 1:1. Then purify it.
