| Hi Boss, Thanks for your interest on my query. Target Peptide is Boc-Leu-Val-Ile-OMe i will couple Boc-Leu-Val-OH + H2N-Ile-Ala-OMe. My problem is, After De-Boc'ing of Boc-Ile-Ala-OMe with Formic Acid,i can remove the formic acid after that how to work-up should takes place with care,those procedure if you provide i will be grateful.becoz sometime i am not getting any yield. Thank you so much |

| i see ,the procedure may be the problem .you can try to use 50%TFA/DCM 1h DeBoc,then evaporate the solution to dryness, then wash the precipitation with 1M sodium carbonate/water solution ,collect the precipitates by filter ,dry the product in desiccator. 6M hydrochloric(gas)/dioxane solution also works well. Formic Acid can react with amimes after you deBOC.it's a side rection . i hope the procedure can help you |
