 Magic Mixture Did anybody use the magic mixture for difficult peptide synthesis? I found the recipe for this mixture in novabiochem cataloge book: DCM/DMF/NMP(1:1:1) with 1%TritonX100 and 2M ethylenecarbonate at 55 centigrade as the solvent system for acylation. I don't know if it works well and worry about the tube will blow out because the bp of DCM is lower than 55 centigrade.        |

| Re: Magic Mixture I never saw any use in magic mixture. I tried several times and it never helped a lot. Choosing a good resin (sequence depending - mainly I would suggest an all-PEG-resin such as ChemMatrix) and a good coupling reagent (e.g. Pybop) usually has a much better effect. Using microwave assisted peptide synthesis helps a lot too (it is our routine method nowadays). All of course depends on the cause of the difficulties ... meaning the sequence ... Andreas |
