| I have taken some PCR products and done another PCR on them, with the same primers but a different Taq. I ran them on a gel, which shows bright bands of the desired size, but with some smearing. Is this because there might simply be too much DNA as a result of the second PCR? Does anyone know if I should avoid using these PCR products for cloning? Thanks |

| when u use A PCR - product as a template for the same reaction , the template may prime to itself creating such smear, so, if u have a good amplification with a smear u can gel - purify it or clean it up and use it for cloning.. |

| I am trying to do a PCR using a PCR product. I used 2 µl as template. When I ran a gel of the PCR, I had a smear. What should I do? |
