Default PCR Agarose Gel Electrophoresis

Hello everyone,

I know running PCRs on agarose gels in common practice. However, are there any tips from anyone to increase the quality of the gels?

I have a few:

For environmentally friendly PCR agarose gels, do not use Ethidium Bromide. Instead, use SYBR orange dye for DNA to stain.

Also, make sure you use a very good gel documentation system. The very bad ones use a digital camera contraption. The best ones have a dedicated gel documentation system that has live previews, is NOT connected to a computer, and prints glossy photos.

I usually re-use my agarose gels 4-5 times, each time with a new buffer. It works very well!

I use SYBR green. Works nicely. Also, the gels spend a very short time on the trans-illuminator

Lightbulb Re: PCR Agarose Gel Electrophoresis

I melt down my agarose gels or dry them, smash them into powder and then re-use the powder as agarose. Only the tricks is - each time after I'm done with the gels, I make sure that the BPB, XC and DNA fragments are all ran out of the gels... That way I'm left with a totally empty gel that can be re-melted or dried and re-weighed
