PCR Primer Dilution
How to Dilute Your PCR Primers EASY
If you get your PCR primers lyophilized (ie dry powder), you can re suspend it to create a 100 uM/L concentration.
How to Prepare PCR PRIMER STOCK
For an amount of 38.5 nM of lyophilized primer provided by the primer supplier, one adds 385 microlitres of double distilled water or Tris buffer (some use TRIS EDTA - BUT note that EDTA will inhibit the PCR reaction due to Mg binding - so better to use TRIS buffer) buffer to get 100 uM/L stock solution.
How to Prepare PCR PRIMER WORKING SOLUTION
To prepare 10 microM/L (or 10 picom/ul) as a PCR working solution, one takes 10 microL stock solution and one adds this to 90 microL of Tris buffer or double distilled water to generate a 10X dilution (thus going from 100 to 10 microM).
