| Hello, i want to use oligonucleotide primers larger than 30-mers ie 45 nucleotide oligonucleotides. This is because i would like to add the restriction site plus a few bp on either side of the cleavage site to make efficient restriction digestion after PCR and cloning is this ok? how should i optimise my sequence? thanks again |
