| Hi everbody I try to evaluate the percentage of iNKT cells among othe CD3+ lymphocytes in my tissue sample by using CD3 and va24 specific primers. generally iNKT cells are less than 1% of CD3 positive cells but as you can see in the attach pics after a pcr amplification (40 cycles) the band for NKT celsl (191 kb) is somehow darker than the band for CD3 lymphocytes (103 kb) while I expect a quite darker band for CD3 because of the higher number of CD3+ cells in my samples in comparison to NKT cells ( 99% vs 1%). would you please help me to find what the problem?
