| I have to do an extended PCR to clone a large DNA fragment. I have tried the more processive Taq enzymes on sale to no avail. I cannot get my band of length 5 kb. I have a feeling that it is the cycling program. Any one have any ideas on how to improve long pcrs? thanks |
