| hi all, I have a question about linear DNA single nucleotide mutation. I have a sequence of GFP. I want to add an NcoRI site in front of the first base so it could conjugate with another promoter I have (which has a NcoRI site at the end). But the GFP I have right now has a internal NcoRI site. I want to mutate it so it is clean inside. What method do you guys think is the best one to do the mutation? Overlapping PCR maybe? I am eager to learn things from you all. Smile Cheers |
