Exclamation Colony PCR Problems

Hello,
I am conducting colony PCR however I get a problem as I do not get any positive results from the experiment!

Does anybody have a good protocol for E.coli colony PCR?
My protocol is:

Pick colony using pipette
Put the pricked colony in 20 ul of milliQ Water.
Heat sample for 8 min (boil)
Use 1 ul of this mix as PCR template and conduct PCR.

Should I use something like an enzyme to extract DNA?

Thanks

Default Re: Colony PCR Problems

when i do colony pcr i pick a colony (small-ish) with an sterile tip and add it directly to the pcr mix... the high temps during pcr will lyse your bugs... it had always worked for me
