How can we perform multiple site directed mutagenesis on the same gene????? plx help me.............

sequentially. Because you don't want to overload your primers with mismatches, the best way to do this is to make a few changes, transform, miniprep, sequence and if everything is as it should be, continue to make additional changes to your newly mutated DNA. Probably more laborious than you hoped but you'll get really good at doing this procedure.
