 Viral Growth Conditions Hi everyone, I have a viral sequence in a pcDNA3.1- backbone. I have a couple of questions. 1) I was wondering if the transformation conditions are particular to pcDNA3.1-, i.e. at 37C, or if I have to grow them as a virus? 2) I've been told that to viral growth conditions are the following: heat shock the cells at 37C, then transform at 30C and grow the cells in LB agar (with the appropriate antibiotic) at 30C. Does this seem right for viral growth conditions? Thank you so much for your time and help! Maria  |
