| I am looking for a good method to isolate large (215 kb DNA) without resorting to old-fashioned CsCl centrifugation. I have tried some of the methods used for BAC/PAC purifications but there must be some trick because I don't get very good DNA. Any suggestions? |

| Hello Malabi, there are many kits for large DNA vector purification. Here is a protocol using gel filtration: SpringerLink - Journal Article Has anyone heard of ProPrep Kits? They are used for large BAC and large plasmid purification. CsCl gradients are still the gold standard I think for large plasmid isolations / purifications. |
