Question 2D Electrophoresis



Hello,
I have some problem in 2DE. My 2D gels are not coming with large number of spots. The spot intensity and no. both are less. And higher molecular wght protein spots are missing. I m doing in 4-7 range with more protein quantity (500 ug).
Pls let someone suggest me what shud I do with it.
And is there any other method of precipitating protein rather than TCA acetone precipitation. As in this method it is tough to dissolve protein completely in rehydration buffer.
