Default Inquiry on COX-2 assay

Hi everyone, i'm doing the research on COX-2 inhibition. Now i'm reviewing the principle of COX-2 detection by colorimetric determination of TMPD. I have some couple of the questions that i'm curious about according to Cayman protocol...
What the role of heme and potassium hydroxide in the COX-2 assay kit?

Thank you for your time and looking forward to the answer
