| Hello, I have been isolating RNA many years already without any problem, but now I have some problem with RNA degradation and I can't solve it. The point is that only tissues from species Apodemus are degradated, the same tissues from mouse or birds are OK using the same kit and protocol. I'm sure that we don't have problem with tissue handling and homogenization proces. We are using Trireagent from Sigma, and one problem is that we are having it in fridge, but on the label is written room temperature. Do you think that this may be the big problem? Probably, tissues from Apodemus have high content of RNAses, do you know some tricks how to handle with such tissues? Is there some difference in effective RNAse inhibition between various products like TriReagent, Trizol, RNAzol or RiboZol? Thanks, Romana |
