Default restriction digests for HIV -1 LAMP products



Hello all,
I am doing my work on LAMP assay for semi quantitative detection of HIV-1. I am in the initial stages of adaptation. I have been having a problem with constantly realizing unspecific amplifications. I would want to do a restriction digest for Integrase and P24 primer binding sites. How do I know the best restriction enzymes to use and how do I identify these sites? Thanks in advance

probably good to look for a site between the F1 and B3 region (target) or maybe the loops. Try NEB Cutter: put sequence in and it will give you sites, different options. I am doing similar work, what are you using to detect your product?
