You are extracting plant gDNA i assume,

go to this site

cilr.uq.edu.au/UserImages/File/Plant%20Genomic%20DNA%20Extraction%20by%20CTAB%20_ 2__Fiona.pdf

Depending on which extraction kit your lab has, i.e from invitrogen or Qiagen follow that specific manufacturers protocol. The one i post above uses Qiagen, scroll down to "Plant Genomic DNA Extraction using Qiagen plant mini kit". Usually the manufacturer does supply you with info necessary to use the kit.

Especially for small amounts of DNA, I have had bad experiences of losing my DNA on Qiagen columns after following their protocol... And, no it's not my technique. Ashu if you want to send me a message, or email I can happily give you pointers on improving your yield off Qiagen columns by modifying the protocol, if you also find that the kit protocol does not work well.
