Can we detect a surface marker (by surface staining) and intracellular cytokine (by permeabilization) for a certain cell population at the same time by flow cytometry?

It should be possible - I have certainly done concurrent extracellular and intracellular stains for flow cytometry before. It tended to work better if I did the extracellular stain first, then permeabilize and do the intracellular stain.

I thank you for your reply cause yesterday one of my friends gave me a reference saying the same. Thanks for sharing your experience.
