Default Can cells amplify and pass the plasmid transfected?

Dear all,

I am currently doing some transfection, and if I do it when the confluence is 50 to 60% I can get more cells that give me the GFP, however, if I wait until higher than 70%, I get less GFP. I am wondering whether the cells can amplify the plasmid that is tranfected into them? And can them pass the plasmid to their offspring when they divide? If not, why do I get less GFP when I tranfect them when the confluence is higher than 70%?

Thanks.

Lei

as u know that the replication of plasmid need energy in 50-60% the cells will get gud energy frome media so it will replicte the plasmid also and in 70% the source of enery will be less so it will not allow the plasmid to rep
