Default G-banding



I am a student. We are having problems getting good slides after digestion with pancreatin. Many different times have been tested but the chromosomes are still staining poorly-too lightly and banding is indistinct.
Also, the chromosome source is our own blood cells, and we used PHA as a mitogen. I did not get a large pellet out of any of my samples, and many of the chromosomes are in a very shortened state. Is there any way I could vary procedure for better results next time?
I appreciate any other comments relating to this-I am just learning.
