Default Ammonium persulfate problems



I dissolved 0.1g of APS in 1ml of distilled/autoclaved water. I then made 100ul aliquots and stored at -20C. The APS solution never froze and when used within a week, 50% of the gels polymerized and no banding occured after destaining in the ones that did polymerize. I've tried two bottles of APS. Are there any suggestions?

Hi

As long as APS is fresh, it's ok. you can keep at 4'c for at least 2-4 weeks, covered with aluminium foil. I don't think we need to keep at -20.
My own way to do short cut (to make sure gel is formed) is that adding APS and Temed a bit more than what the standard protocol says.
I am not sure what banding means. If you mean protein band, it's nothing to do with APS. it's your protein quality and quantity.
