Default Shrimp Alkaline Phosphatase - Biology and Protocol

Shrimp Alkaline Phosphatase ( SAP ) catalyzes the dephosphorylation of 5’ phosphates from nucleic acid such as DNA or RNA.

Unlike calf intestinal phosphatase ( CIP ), SAP activty is inactivated by heat treatment for 15 min at 65°C. Thus the
whole proceeding including restriction enzyme digestion, dephosphorylation,
enzyme inactivation, ligation or 5’ end-labeling can be performed
in one single tube by just adding the appropriate reagents.


SAP protocol:
  • DNA > 1 μl
  • 10X SAP Reaction Buffer 2 μl
  • Water, Nuclease-Free up to 19 μl
  • Shrimp Alkaline Phosphatase (1 unit/μl) 1 μl
Incubate 37°C for 30-60 minutes

Inactivate SAP by heating 65°C for 15 minutes
