| I am growing up chemically competent E.coli cells for cloning. I have one clone that grows on an LB+Amp plate, but when I inoculate a liquid LB+Amp culture (incubating over night shaking at 37) with this strain I get no growth. I thought that maybe my first Amp plate was faulty so I re-streaked the culture on a new plate and it grew just fine. I tried growing it in liquid culture again to no avail. I have many other strains I have done this with so I'm fairly confident that there is nothing wrong with my protocol. This is the only strain I am having trouble with. Any suggestions? |
