Growing Staph.aureus

Hi, I really need your help group, I have Staph.aureus in preservation culture and I am trying to grow it in M9 minimal media (liquid) but unfortunately they are not
growing
I tried different volumes ex: 5ml M9 media + 200ul S.aureus (preservation culture) then leave it in shaker incubator @ 37c over night.
has anyone had any experience growing them ?

What do you mean by "preservation culture"?

What I mean by "preservation culture" is that I grew the bacteria in LB media then made a preservation of it by adding 50% of the LB(bacteria) culture + 50% glycerol then kept it in -80c freezer.

I'd run a control with LB medium to confirm you can recover stpah from the preservation culture. If you ca, it would appear your minimal medium does not suport grwoth of the strain you you have.

Why are you trying to grow S.aureus in M9 minimal media? Isn't it easier to just grow it in Nutrient Broth or grow it on (a selective) agar and then take some material and store it in the freezer?
I've googled the M9 minimal media and i found that its used for E.coli only. Are you trying to modify this media to let S.aureus too?
