antibiotic production test

hi...2 all..

i hav to see the test "effect of antibiotic (produced by fungi) on the bacterial poured plate..so due to antibiotic disc it shud show a resistancial colony surronding this..bt i hav never get the result perfactly...
why..? is there ny other way 2 see test accurately.....
help me...

it is done by spreading on plate nt by streaking. so what you have to do is to taka media plate L.B Agar or nutrient agar and spread you bacterial culture on it so that it produce a lawn on the agar surface. after spreading attach the disc on the surface at different position and then incubate. and on next day if it is sensitive then there will be no growth oround disc otherwise if resistant then growth will be there.

i hav told that i hav done this expriment as ur telling..bt somtimes i got a small area that shows resistance...not a perfect colony i hav seen....
thats why i m asking wat may be the factors...due to which i didnt get...
is there ny other way 2 do this...
if i tak agar plate n spread bacterial suspension over it.. n then we put the penicillium inoculum at the centre of plate...wud it shws resistance against bacteria wen it will produces antibiotic....
