Default peptide mass fingerprinting to identify antibodies!



Is it possible to identify an antibody IgG1 by tryptic digestion and PMF database searching?.

Depends. I have run serum and saliva on MALDI-TOF for PMF, and seen antibodies show up. Usually the list reads out the type of immunoglobin and species (obviously not the corresponding antigen for the antibody).

But if you have a standard (100% known Ab) and want to test an unknown to see if they match--you might be able to compare spectra (PMF). The purity of the Abs in this case would be very important as any other proteins in there will also show up.
