| Our lab has been using an inducible deletion model for several years with no problems. Transgenic mice expressing the Cre-recombinase gene under control of the interferon inducible promoter Mx1 (MxCre) recombine floxed genes after treatment with IP injections of polyI:C. In the last month we have experienced 50% deaths from the polyI:C injections. SIGMA suggested that there was endotoxin in that particular lot as they do not test for endotoxin and the polyI:C in not intended for in vivo use. BUT we have not had any problems in the past. Have we just been lucky for the past two years and gotten cleaner batches of polyI:C? Has anyone else had similar problems? Thanks, Frustrated graduate student ! |
