Default Y2H Identification of Prey Protein



Hi all,
It is my first post. Stuck at yeast two hybrid system. Screening is done. Unable to identify the target protein. Isolated yeast plasmid DNA , electroporated in E. coli ans selected for Amp resistance (prey vector). Some colony appeared. Purified the DNA from E. coli BUTTTTT the digestion profile does not give the desired match.
Can anyone suggest something.
Ur kind help is required.
