| Hi everyone! I'm pretty much a beginner scientist, but I'll try to define my problem as clear as I can: I'm working with rat brain (hypothalamus), and I've been trying to isolate the leptin receptor, which is a plasma membrane protein, so i could analyse it by western blot. Most times, the only bands I could detect were nonspecific (I could see specific bands a few times, but they were lighter than the nonspecific ones). It might be the quality of the antibody, but I think that it could also be the result of my not knowing the right way to isolate a membrane receptor from the brain tissue-using different preps gave me different quality of the specific bands. Any advice, anyone? Thanks , and have been trying to isolate and prepare a membrane protein |
