| Hi, I am just starting to work with a recombinant virus and hoping a few questions can be answered. My adenovirus has a titer of 10*12 VP/ML. Does this mean to acieve an MOI of for example one I need to dilute 1ul into 1ml (reduce to 10*9) of media and add 1ul of that to a cell culture dish containing 1 million cells. Also, if my virus has been purified and the titer is done using OD then my 10*12 viral particles/ml represents both live and dead virus. In general, can I assume most is live? Otherwise how would I determine an MOI? Thanks. |
