Ebola Antigen-capture ELISA : monoclonal antibodies v.s recombinant nucleoprotein (rNP) of Ebola virus subtype Zaire is coated on microwell immunoplates (BioHit) at a concentration of 100 ng/well in 100 l of PBS for 2 h at room temperature (RT) or overnight at 4C. After washing the plate three times with PBS containing 0.05% Tween 20 (PBS-T), and 200 l of PBS-T containing 5% nonfat milk (milk–PBS-T) are added to the wells for blocking for 1 h at 37C, or blocking with 200 l of 5% nonfat milk in PBS for 1 h at RT. After the plates are washed with PBS containing o less 0.05% Tween 20.
Procedure
Dispense in well 100 l of control containing rNP or clinical specimens diluted in dilution buffer sample (phosphate-buffered saline [PBS] containing 0.05% Tween 20, 1% Triton X-100, and 5% nonfat milk) and incubate plate for 1 h at RT. The plates are then washed with PBST, and 100 l of rabbit polyclonal antibody anti rNP diluted 1:1000 in 0.5% nonfat milk was added to each well. After 1 h of incubation at RT, the plates are again washed with PBST and HRPO-labeled anti-rabbit IgG (Dako) are added. The plates are incubated for 1 h at RT. After another extensive wash with PBST, 100 l of ABTS substrate solution are added and the Abs is measured at 405 nm after 30 min of incubation at RT.

Research IgG vs Ebola virus with ELISA method : dispense in polyvinyl chloride microtiter plate (BioHit), 100 l of antigens for EBO-R, or EBO-S, or EBO-Z diluted 1:1000 in PBS, and adsorb at the microtiter plate overnight at 4 C. Remove antigen from the wells washing 3 times with PBS-T.
Procedure
Sera are diluted 1:100 and 4-fold through 1:6400 in 5% nonfat milk in PBS-T and allowed to react with the antigen-coated wells. Bound IgG was detected with mouse anti-human IgG (g-chain specific) conjugated to horseradish peroxidase (Dako). Wash the wells 3 times with PBS-T and dispense 100 l of TMB substrate. Incubate for 15-20 min. and stop reaction dispensing 50 l of 2 N H2SO4 Read the Abs at 450 nm.
