Default How can I improve cell numbers for sorting???



Im having troubles getting good Treg cell numbers after sorting, and have found that Im actually losing a lot of cells in the labelling process (3 color indirect). I compared tipping the facs tubes after washing to using suction and I still lose about 50% of my cells. This is not usually a problem for profiling, but we are trying to sort for Tregs that are only 1% of cells sorted. How can I maximize my cell numbers between labelling and sorting?
Any suggestions are greatly appreciated.
