Default Quantification?!



Dear all,

how do you quantify or standardise your FACS results? I know there are several forms of quantification beads, e.g. Quantibrites from BD for this purpose. But as I am using the same FACS-machine all the times and make sure that the voltage for the channel I´m looking at is constant, e.g. 600 for APC-Cy7, do I really need this???

Bjorn,

While compensation beads are not ABSOLUTELY necessary, there is an entire built in compensatory system that can be used with the beads. This will make your data cleaner and with far less error, as well as just simply more correct. There are many factors other than just the voltages that can throw off the results, including blockage, lower pressure, laser/filter changes or maladjustments, etc.

Get the beads if you are having problems with the data and you have some extra money. Dont get them if your data already looks clean with little background.
