Default Isotype control - substraction or division

Hello to you all.

I encountered some difficulties running a flow cytometry analysis using the MFI values of the specific values against the isotype-matched control, as I haven't been able to find an unambigous answer as to whether the isotype value should be divided by or subtracted from the specific antibody value. I tried both methods, and while division yielded a very interesting and statistically significant pattern, substraction results an a highly-variant (nonsignificant) distribution of values.

Can anyone clarify which method - division or substraction - is correct to use?

Thanks
