| Hi, i am going to to analyse some Corynebacterium cells which are GFP tagged with a gene in the genome using FACS. The thing is i am not sure how to prepare cells for FACS. Does anyone have a good protocol? Does one harvest the cells and wash them with PBS 3 times and then store them in the 4 degree fridge until FACS is done? Also how long can cells be stored and what should be the concentration of the cells for FACS? Also how much cell sample is needed to conduct FACS? At what steps should cell counting should be performed before FACS or just after washing step? thanks |
