Question I have problem in PBMC isolation


I couldn't get nice images on flowcytometry when I isolate PBMC with Ficoll.Is it requisite for me to use NH4Cl to split Rbc?

hi, you can use ACK lysing buffer to lyse red blood cells and directly stain with antibody and run flow. It is not necessary to use ficoll to purify the lymphocytes.
