Previous work by K. Prasadan showed Morpholinos enter rodent embryonic pancreatic cells from the extracellular space. Several papers from University Paris Diderot-CNRS show uptake into the pancreas of rat pups after s.c. injection of bare or fluor-tagged Morpholinos:
Uzan B, Figeac F, Portha B, Movassat J. Mechanisms of KGF Mediated Signaling in Pancreatic Duct Cell Proliferation and Differentiation. PLoS ONE. 2009;4(3):e4734. Epub 2009 Mar 6.
Figeac F, Uzan B, Faro M, Chelali N, Portha B, Movassat J. Neonatal growth and regeneration of {beta} cells are regulated by the Wnt/{beta}-catenin signaling in normal and diabetic rats. Am J Physiol Endocrinol Metab. 2009 Nov 17. [Epub ahead of print]
While we at Gene Tools are habitually skeptical of claims of delivery of bare Morpholinos dosed systemically in vivo, this is a case where previous careful work has shown uptake of bare oligos into embryonic cells of this type. We anticipate further work in this system with keen interest.

Hi,
I am trying to insert the gateway cassette (according to Invitrogen instructions from their kit) into pGL3basic to receive a gateway compatible luciferase reporter vector that is promoterless. Unfortunately, I cannot get any colonies ever. I am pretty frustrated and under dealine pressure... any suggestions? HEEEEEEEEEEELP!
