| Dear all, I have got a problem from my MSP? I think, it may be caused from specific primer degradation. Anyone ever experienced this trouble? Please, give me the suggest. I'm so blue .I tried to perform the MSP using the same PCR master mix (fresly prepared before running in the thermal machine). Everything is the same (PCR reagents and bisufite modified DNA), except the working primers. I used the different working primers (diluted in the different time by 1xTE buffer) but they are from the same stock primer. I found the specific product from first working primer but not found from other lots of working primers. I have done several time of MSP and with many lots of working primers to find out the problem. I assumed that MSP fail from Primer Degradation....?? How long can we stored the stock primer at -20 C? What about thawing conditions of primers? How can we check the degraded primers?
