Sample dilution
 
Hi everyone,

I'm a newbie with ELISA and I have a basic question.

I'm working with mouse serum in order to quantify IFN-gamma and my supervisor asked me to dilute 2x the sample in assay diluent.

So once I get my OD, in order to calculate the original concentration :

Do I have to multiply the OD by 2, or do I have to multiply the calculated concentration by 2?

multiply the calculated concentration off the standard curve by 2. Generally you dilute so that it will be in the OD range for the standard curve
