Default DNA restriction digeestion

HI,

I have a basic question, does residual RNA disturb a plasmid DNA restriction digstion. Can i skip the RNA removal step while plasmid isolation.

Thanks

Hello Tuttu,

RNA does not really impact on DNA restriction digestion, although I would remove the RNA by RNases. Usually RnaseA is already added to the lysis buffer so it would not be extra work.

Welcome to the forum!
please tell us how it worked out...
