Default agarose gel help



Does genomic DNA absolutely need to be cut even if only one band is necessary to be visualized in the gel? I have tried running genomic DNA but no bands appear and DNA doesnt show up as if it has stayed in the wells. any ideas as to my problem?

Hi

what type of genomic DNA you are going to isolate? as there are different procedure for different organisms.
Usually when we isolate genomic DNA by routine procedure it is sheard off into fragments of around 20-25 Kb and when you run it on agarose gel, you get a single band.
try to load lesser amount of DNA on your gel and also prepare 0.6 or 0.8% agarose Gel then hopefully you will get band for genomic DNA.

it is genomic dna from wheat germ. about how much DNA should i load and how should i go about shearing the DNA?

well you can quantify the DNA by using spectrophotometer at 260nm and load in range of 500 ng to 5 ug, then you will have good idea that how much you should load......

i did not work on this type of genomic DNA but i am sure when you prepare Genomic DNA it already get shear and you get a band around 20-25 Kb.....

Run on low agarose gel and i do hope it will be okay..........
